in+一段时间表示将来时
如:I'll come back in a week
in＋一段时间也可用于过去时，意思是”．．．内”
如:he came in two days.
而after＋一段时间通常只能用于过去时，不能用于将来时
如：he came back after a week

in 一般表示从现在起过……时间，用于现在式
after “在……之后”一般用于过去式

in一般用在将来；而after一般用在过去的时间之后。比如：It's two o'clock. I'll come in an hour.(= at three o'olock) / After a quarter after ten he returned home.

此外前者还表示“在某段时间之内”的意思
