两者都对，但是有细微差别，给你举例你仔细体会一下就能明白。
I'm not sure(that) I know how to get there.
I'm not sure(if) I'm pronouncing this word correctly.
第一句：觉得自己知道，但是把握不大（有时候多少含有希望自己没弄错的意思在里面，有时候是直接表示怀疑，给另外一总存在留下的空间很小）。
第二句：基本上是毫无把握。
确实，be not sure 用在口语中更常见，往往省略that。
